贯叶连翘无菌苗茎尖诱导变异体研究

摘    要:以研究贯叶连翘离体诱变及筛选变异体为目的,用贯叶连翘无菌苗茎尖为供试材料,研究了平阳霉素处理浓度、时间、外源添加物对供试茎尖存活率以及激素配比对茎尖不定芽诱导率的影响。结果表明:平阳霉素80mg/L的20min处理可作为适宜诱变处理,添加椰汁有利于茎尖存活;6-BA0.5mg/L、NAA0.01mg/L的激素组合有利于诱导正常型不定芽和获得较高的诱导率。通过研究,建立了贯叶连翘变异体诱导技术程序,获得了金丝桃素含量提高的变异体材料。

Abstract:The purpose of this research is to induction and selection variants of Hypericum perforatum L. Using stem apexes of Hypericum perforatum as material, effects of Pingyangmycin (PYM) concentration and treating time as well as supplementary materials in medium on survival rate of stem apexes and influence of hormone combinations on induction rate of adventitious-buds from stem apexes were investigated. The results showed that the treatment of 80mg/L PYM for 20min was suitable for mutation. The survival rate of stem apexes could be improved by adding CW in medium and the combination of hormones 0.5mg/L BA with 0.01mg/L NAA was suitable for inducing normal adventitious buds and obtaining higher induction rate. The procedure for inducing and selecting variants from stem apexes of Hypericum perforatum was established, and a variant with higher hypericin was obtained.
