益生芽孢杆菌B.licheniformis29质粒pBL29抗性基因的筛选

摘    要:采用改良碱裂解法对30株益生芽孢杆菌进行质粒抽提,仅从1株地衣芽孢杆菌B.licheniformis29菌株中抽提得到质粒pBL29,纯化后通过琼脂糖凝胶电泳观察,确证质粒pBL29为闭合环状、分子量3.7kb左右。通过药敏试验、吖啶橙消除试验、十二烷基磺酸钠消除试验及原生质体转化试验证明:B.licheniformis29菌株具有卡那霉素抗性。而且卡那霉素抗性基因是质粒编码而不是染色体编码。

Abstract:The presence of plasmid in Bacillus was isolated by improved alkaline lysis. Only one plasmid pBL29 was detected in B. Iicheniformis29. Puried and identified by gel electrophoresis, the plasmid pBL29 is cccDNA and the molecular weight'of the plasmid pBL29 was estimated to be about 3.7kb. The sensitivity of Bacillus Iicheniformis29 to 18 antibiotics was determined by inhibitory zone with filter paper. The result proves that B. Iicheniformis29 is resistant to Kan and sensitive to the other antibiotics. The elimination of plasmid by acridine orange or by SDS and the transformation of protoplast indicated that Kan resistance genes are located on the plasmid pBL29, not on the chromosome.
