家蚕HSP20·8基因体外表达及荧光原位杂交研究(英文)

摘    要:前期研究发现,相对于其它组织器官,有一种低分子量热激蛋白在家蚕肾型卵中高量表达,并命名为低分子量热激蛋白HSP20.8。为了进一步阐明家蚕低分子量热激蛋白HSP20.8的基因结构、表达特性及其在家蚕染色体上的分布,根据HSP20.8基因cDNA序列设计特异引物,进行了克隆、表达及荧光原位杂交研究。结果表明,该基因含有1个561碱基对的开放阅读框(open reading frame,ORF),与cDNA序列比较分析发现该基因无内含子序列。重组蛋白的分子量在20 kD左右。热激蛋白基因HSP20.8在家蚕基因组中为单拷贝基因,其位点在染色体近中部区域。

Abstract:Based on the previous research,one gene of Bombyx mori named as HSP20.8 can be over expressed in kidney-shaped eggs than in other organs.In order to get fully understanding of the small heat shock protein gene HSP20.8 such as gene structure,the character of protein expression and gene location in the whole genome of B.mori,the HSP20.8 gene was cloned and expressed and in situ hybridization was carried out.The results showed that the ORF of the HSP20.8 gene is 561 base pairs without introns and its translation product is about 20 kD.It is a monocopy gene in the genome of Bombyx mori and locates at near the middle site of the chromosome.
