落葵的组织培养与植株再生

摘    要:以落葵子叶和下胚轴为外植体,在MS基本培养基中添加不同浓度激素培养。结果表明,下胚轴经1-4mg/l6-BA,2mg/l6-BA+0.1-2.0mg/l2,4-D激素处理,可产生愈伤组织,而子叶却不能。下胚轴愈伤组织在2mg/l6-BA+0.1mg/lNAA处理下芽分化最好。在芽的继代培养中,4.0mg/l6-BA+0.1mg/lNAA处理最佳,平均每个外植体可分化4个芽。在1/2MS+0.3mg/lNAA培养基中生根质量高。再生小植株成活率达90%左右。

Abstract:The cotyledon and hypocotyl for explant of Basella alba L. were cultured in MS media supplemented with various plant growth regulators at different concentrations. The results indicated that hypocotyl produced calli when cultured in mediacontaining 1-4 mg/l 6-BA, 2 mg/l 6-BA + 0.1-2.0mg/l NAA, or2 mg/l 6-BA + 0.5 - 2.0 mg/l 2,4-D, but cotyledon failed. The bud regenerated very well from callus of hypocotyl in presence of 2 mg/l 6-BA + 0.1 mg/l NAA. The bud gave the best rege.neration with4 regenerated shoots fxom each explant of Basella in average when subcultured in a combination of 2 mg/l 6-BA + 0.3 mg/l NAA. The best rooting of regenerated bud was achieved in a combination of 1/2 MS + 0.3 mg/l NAA. The regenerated plantlets reached a survival rate of 90% when transplanted into pots.
