摘    要:以马铃薯优良四倍体栽培种‘唐113’、‘陇薯3号’和‘甘农薯1号’为材料,对不同发育时期的花粉进行了离体悬浮培养.结果表明,培养前将材料于4~7℃低温下处理7 d或0.3 mol.L-1甘露醇中饥饿处理3 d均取得了较好的培养效果,花粉分裂频率分别达到13.2%和13.6%.马铃薯单核期花粉具有孢子体发育的巨大潜力,分裂频率高于其他发育时期的花粉,其中‘唐113’脱分化率达4.1%,是马铃薯花粉诱导愈伤组织和再生植株培养的适宜时期;不同品种花粉离体的脱分化频率表明马铃薯花粉培养存在基因性依赖.

Abstract:Three potato varieties,'Tang 113','Long Shu No. 3' and 'Gannong Shu No. L',were em-ployed as raw material for pollen culture in vitro. The results indicated that pretreatment at 4~7 ℃for 7 days or mannitol at 0. 3 mol ·L~(-1) for 3 days enhanced the division frequency of pollen. The division fre-quency reached up to 13. 2 % and 13. 6 %. Pollen in monokaryotic period had a better potential for develop-ment of microspore,and had a higher division frequency. Pollen culture in vitro of different variety indica-ted that dedifferentiation frequency referred genotype of potato cultivar.
