金针菇粉孢子的产生及核相观察

摘    要:采用插片培养的方法培养金针菇单核菌丝体和双核菌丝体,在普通光镜和扫描电镜下观察单核菌丝和双核菌丝产生粉孢子的方式及粉孢子的形态。通过荧光染料和石碳酸-品红染色观察表明,单核菌丝产生的粉孢子都只含一个细胞核,双核菌丝产生的粉孢子也均为单核。本研究未观察到双核或多核的粉孢子。

Abstract:The monokaryons and dikaryons hyphae of Flammulina velutipes were cultured with insert galss cover. The occurring ways and shapes of oidia produced by mono and dikaryotic hyphae were observed under optical microscope and scanning electron microscope. It was found that single and di nuclear hyphae produced the oidia containning one nuclear only. However the di-and multinuclear oidia were not observed by staining of fluorsecent dye (Bisbenzimide Hoechst 33258) and ordinary dye (carbolic acid-fuchsine).
