小麦种子萌发期水分胁迫诱导表达差异cDNA的研究

摘    要:以小麦幼芽为材料,采用mRNA差异显示方法和银染技术,对经过用16%(-0.5 MPa)PEG-6000溶液处理不同时间而诱导表达的小麦基因进行分离,共得到cDNA差异片段16条。其中4条差异条带呈表达减弱,其余差异条带均呈表达增强。测序结果与GenBank数据库比对,2个cDNA片段与已知基因同源,WSI241与编码胚后期丰富蛋白的Em基因同源性达100%,WSI208与ARA-1 mRNA同源性达88%。大部分都是功能未知的EST序列。其中WSI483(454 bp)与小麦干旱胁迫幼苗cDNA同源性达100%;WSI232与小麦干旱胁迫叶cDNA同源性达100%。WSI301与小麦盐胁迫芽鞘cDNA同源性达98%,WSI267与小麦热胁迫旗叶cDNA同源性达93%,WSI289与小麦ABA处理胚cDNA同源性达92%,WSI268与镰刀菌感染的小麦穗同源cDNA同源性达97%。经反向Northern验证,有6个cDNA呈阳性。可用探针或设计引物作进一步研究。

Abstract:Adopting both mRNA differential display and silver staining techniques to isolate wheat genes induced by 16%(-0.5 MPa)PEG-6000,total 16 differentially expressed cDNA fragments were obtained.Through sequencing and then querying the database of GenBank,two fragments,WSI241(water-stressed induced) and WSI208 were highly homologous to the known genes,but the others belonged to ESTs with unknow function.Among them,the homologous of WSI241 to Em gene was 100%,WSI208 to ARA-1 mRNA was 100%.WSI483 to drought-stressed wheat seedling cDNA was 100%,WSI301 to salt-stressed wheat leaf cDNA was 98%;WS267 to heatstressed wheat flag leaf cDNA was 93%,WSI289 to Wheat ABA-treated embryo cDNA was 92%,WSI268 to Wheat Fusarium graminearum infected spike cDNA was 97%.After further certified with reverse northern dot blot,six fragments proved to be positive,can be used as probe or selected primer to do further research.
