氧对提灯变形鞭毛虫生长及生理特性的影响

摘    要:提灯变形鞭毛虫Psalteriomonas lanterna的生活史分为鞭毛期和阿米巴期两个阶段,其在低氧(体积分数为1.0%)的混合培养液中生长良好;生活在体积分数为1.0%~2.5%O2的培养液中(每周更换2次O2),其细胞内共生甲烷菌消失,变成阿米巴期。将甲烷菌加入失去内共生菌的鞭毛期培养液后,可获得较高的细胞密度。用电镜检查提灯变形鞭毛虫的细胞质,未发现有内共生甲烷菌,该菌仅出现在食物泡中。培养液中加入体积分数为2.5%的O2和0.1mmol/L的甲烷抑制剂(Bromoethanesulfonic acid,简称BrES),能抑制提灯变形鞭毛虫的生长和CH4的生成,同时提高H2的含量;高浓度(10.0mmol/L)的BrES对CH4生成的抑制率为100%,此浓度下细胞停止生长,5d内全部死亡。阿米巴期对氧和BrES的忍受力高于鞭毛期。用细胞化学染色方法检查与氢化酶体和线粒体相关酶的活性表明,提灯变形鞭毛虫缺少细胞色素氧化酶和过氧化氢酶,含有超氧化歧化酶。

Abstract:Amoeboflagellate Psalteriomonas lanterna tolerated low oxygen tensions. When cultured with 1% O\-2, the organism grew quickly.When this organism was cultured in a medium containing less than 2.5% of oxygen (refreshment of the oxygenated gas phase twice a week), its endosymbionts disappeared and changed into symbiont-free flagellates. When the symbiont-free flagellate was co-cultured with strain of Methanobacterium formicium, the high density of flagellates was presented in the medium. Evaluation of methane production showed that methanogenic consumed hydrogen.The ultrastructure of cells showed that methanogens were also present in the food vacuoles but were never found in the cytoplasm. Growth of flagellate with endosybionts and methane excretion were inhibited at concentrations of 2.5% oxygen and 0.1 mmol/L BrES,but H\-2 accumulated increasing. The inhibition rate reached to 100% and the cells stopped growing as well as the cells died at concentration of 10 mmol/L BrES. Amoebae tolerated higher both of oxygen and BrES tensions than flagelles.Both functions of hydrogensome and mitochondria were observed with cytochemical staining method. It was shown that both of catalase and cytochrome oxidase could not be found in the cells, whereas superoxide dismutase was presented.
