牛IgG轻链的分离纯化及其多克隆抗血清的制备

摘    要:实验应用凝胶过滤和盐析等方法直接从牛初乳中分离纯化IgG,并应用化学方法断裂回收IgG轻链。分离纯化获得分子量约27.66 ku的IgG轻链蛋白溶液,蛋白质含量为0.105 mg.L-1,浓缩后免疫家兔,获得抗轻链抗血清,效价>11∶6,免疫双扩散和免疫电泳均为特异性条带,表明得到了纯化的IgG轻链及其特异性抗血清。

Abstract:The IgG was isolated form the bovine colostrums by the chromatography and salt fraction method,and apply the chemistry method to split to recover the IgG light chain.300 mL protein solution was obtained and the concentration of the purified protein was 0.105 g·L-1.Its molecular weight(M.W) was proved to 27.66 ku by SDS-PAGE.Rabbits were immunized with the concentrated protein solution and the rabbit antiserum to light chain of IgG was obtained.Double diffusion shown the titre >1∶16.The related application of the antiserum is in the research now.
