芙蓉鲫(芙蓉鲤♀×红鲫♂)及其原始亲本间的遗传关系

摘    要:利用RAPD和微卫星标记技术,对芙蓉鲫及其原始亲本(红鲫、芙蓉鲤、散鳞镜鲤、兴国红鲤)5个群体的遗传关系进行研究。利用20条随机引物进行RAPD分析,其中12条在5个群体中均能扩增出特异条带。统计分析显示:芙蓉鲫与红鲫、芙蓉鲤、散鳞镜鲤、兴国红鲤的遗传相似性分别为0.8757、0.7478、0.7419、0.7449,遗传距离分别为0.1327、0.2906、0.2985、0.2944。在进行微卫星标记分析时,所扩增的29对微卫星引物中共有13对在5个群体中均能扩增出特异条带。统计分析显示,芙蓉鲫与红鲫、芙蓉鲤、散鳞镜鲤、兴国红鲤的遗传相似性分别为0.8717、0.7434、0.6680、0.7552,遗传距离分别为0.1374、0.2965、0.4034、0.2808。两种分析结果均表明,芙蓉鲫的遗传结构更接近于父本红鲫,与外祖父兴国红鲤的遗传相似性比与外祖母散鳞镜鲤的遗传相似性要大,其遗传结构更偏向于具红色体征的原始亲本。

Abstract:Furong crucian carp(Furong carp♀×red crucian carp♂) is a hybrid that combined with remote hybridization between carp and crucian carp and close crossing among breeds of carp.In original brood stocks of Furong crucian carp(red crucian carp,Furong carp,scattered scale mirror carp and Xingguo red carp),two red stocks were used.Previous research suggested that there was a heterosis when red brood stock was used in hybridization between carp and crucian carp.The present dealt with the genetic relationship between Furong crucian carp and the four original brood stocks by random amplified polymorphic DNA(RAPD) and microsatellite DNA methods,and the genetic effect of red stocks was discussed.In total 20 RAPD primers,it was found that 12 primers amplified the proper DNA bands in five populations of fish.The genetic identities of Furong crucian carp and red crucian carp were found to be 0.8757,Furong carp 0.7478,scattered scale mirror carp 0.7419 and Xingguo red carp 0.7449,and the genetic distances were 0.1327,0.2906,0.2985,0.2944,respectively,based on the RAPD bands.In total 29 microsatellite primers,there were 13 primers that amplified the proper DNA bands in five populations of fish.The microsatellite bands analysis showed that the genetic identities of Furong crucian carp and red crucian carp were found to be 0.8717,Furong carp 0.7434,scattered mirror carp 0.6680 and Xingguo red carp 0.7552 and,the genetic distances were found to be 0.1374,0.2965,0.4034,and 0.2808,respectively.It can be conclusion that the genetic structure of Furong crucian carp was biased to the male(red crucian carp) and grandfather(Xingguo red carp),indicating consistent with the theory that there is a close relation ship between genetic basis of the red broodstock and heterosis.
