结球甘蓝的组织培养与快繁研究

摘    要:目的]研究适合结球甘蓝组织培养与快速繁殖的最佳方法与培养基种类。方法]以结球甘蓝无菌苗的胚轴和子叶为材料,以MS为诱导和增殖的基本培养基,以1/2MS为生根基本培养基,比较不同浓度及配比的生长调节物质对增殖及防止玻璃化苗的影响。结果]接种于诱导培养基MS+6-BA 3.00 mg/L+NAA 0.10 mg/L中的子叶可长出愈伤组织,胚轴可直接分化不定芽;在增殖培养基MS+6-BA 1.00 mg/L+NAA 0.10 mg/L上生长的组培苗生长势强,玻璃化苗数最少,增殖率最高,平均增殖系数为3.8;接种于生根培养基1/2MS+NAA 0.10 mg/L和1/2MS+IBA 0.20 mg/L上的组培苗生根率最高,达100%。结论]最佳诱导培养基为MS+6-BA3.00 mg/L+NAA 0.10 mg/L;最佳增殖培养基为MS+6-BA 1.00 mg/L+NAA 0.10 mg/L;最佳生根培养基为1/2MS+NAA 0.10 mg/L和1/2MS+IBA 0.20 mg/L。
