氟苯尼考在鲟鱼体内的药物代谢动力学研究

摘    要:采用高效液相色谱检测组织中药物含量,研究水温在(20±1)℃时,杂交鲟经口一次性给予10 mg/kg氟苯尼考后在血液、肌肉、肝脏中的代谢规律.试验结果表明,氟苯尼考经口给药后在鲟鱼血液中的代谢属于一级吸收一室模型,在肌肉和肝脏中属于一级吸收二室模型,且分布广泛,代谢迅速.主要药代动力学参数如下:血液、肌肉、肝脏中的达峰...

Abstract:The pharmacokinetics of florfenicol were studied in blood,muscle and liver in hybrid(Acipenser schrenckii×Huso dauricus) sturgeon sampled after intravenous injection at single dose of 10 mg/kg body weight by HPLC,and the data were analyzed by pharmacokinetics computer program 3P97.The results showed that the concentration-time course of the florfenicol was described by one-compartment model with 1st order absorption in the blood and two-compartment model with 1st order absorption in the muscle and liver,with wide distribution and rapid metabolism.The main pharmacokinetics parameters in blood,muscle and liver were found as following:Cmax 6.43,5.81,and 6.062 μg/ml;Tmax 9.84,8.88,and 0.51 h;t1/2α29,4.61,and 0.27 h;t1/2β 9.45,21.69,and 11.04 h;AUC 169.02,227.41,and 50.56 mg/(L·h);V/f 0.81,1.07,and 0.80;CL/f(s) 0.059,0.051,and 0.2 L/kg;recovery≥90%,R2=0.9996.
