中草药提取物MACC5对大鼠抗Thy1.1系膜增生性肾炎模型的疗效观察

摘    要:目的观察中草药提取物MACC5(为广东医学院肾脏病研究所提取物代号)对SD大鼠抗Thy1.1系膜增生性肾炎实验模型的疗效。方法制备兔抗大鼠胸腺细胞免疫血清(ATS),再将ATS经尾静脉注射给SD大鼠,诱导其发生系膜增生性肾炎(MsPGN)后,随机分为3组,每组5只;试验组分别经腹腔注射MACC51.0mL和3.0mL,对照组注射生理盐水(NS)1.0mL;每日观察其尿量、尿蛋白定量,并于第11天处死各大鼠,取肾组织切片观察系膜增殖程度,并用免疫组化法检测肾小球系膜区细胞增殖核抗原(PCNA)的表达。结果两试验组24h尿量均多于对照组(P<0.05);两试验组尿蛋白定量均少于对照组(P<0.01);两试验组单位肾小球系膜细胞(GMC)数均少于对照组(P<0.001); MACC53.0mL试验组肾小球系膜区PCNA的表达低于对照组(P<0.05)。结论MACC5对大鼠MsPGN实验模型具有利尿、降低尿蛋白和促进肾小球系膜损伤修复的作用。

Abstract:Objective:To observe the effect of MACC5 isolated from traditional Chinese medicine on the rat with mesangial proliferative glomerulonephritis(MsPGN)induced by anti-Thy1.1 antibody.Methods:Anti-Thy1.1 serum(ATS)was produced,and then injected intravenously to SD rats in order to induce experimental model with MsPGN.The rats with MsPGN were randomly divided into 3 groups,and each group contained 5 rats.Both of experimental groups were injected peritoneally with MACC5 1.0 mL and 3.0 mL,respectively,whereas control group received a peritoneal injection of normal saline 1.0 mL .Urine volume and urinary protein were examined everyday. The rat was killed on the 11th day,and the expression of PCNA in the glomeruli was detected by immunohistochemistry method.Results:The urine volume of experimental groups was more than that of control group (P<0.05),the urinary protein and the amount of GMC of experimental groups were significantly lower than those in control group (P<0.01-0.001),and there existed a difference in the expression of PCNA between MACC5 3.0 mL group and control group (P<0.05).Conclusion:It is suggested that MACC5 possesses effects of diuresis and reduction of urinary protein excretion,and promotes the repairing process of mesangial lesions of MsPGN.
