摘    要:目的]从分子水平上探讨四川4个黑山羊品种的遗传多样性和亲缘关系,为黑山羊品种的保护及合理利用提供理论依据。方法]采用mtDNA多态性标记对四川4个黑山羊品种D-loop序列多态性进行分析。结果]供试黑山羊品种mtDNAD-loop片段长度为1210~1212bp,共检测到5种单倍型,群体间共有多态位点65个,单一多态位点33个,简约信息位点32个,平均核苷酸歧异度为0.001518,D-loop序列多态性较贫乏;经聚类分析,乐至黑山羊和金堂黑山羊首先聚在一起,后与建昌黑山羊聚合,最后和自贡黑山羊聚在一起。结论]4个黑山羊品种的mtDNA遗传多样性贫乏,分化程度低;聚类分析结果与品种来源和生态地理分布相一致。

Genetic Diversity of mtDNA D-loop of Different Black Goat Breeds in Sichuan Province

Abstract:Objective] To explore genetic diversity and relationship from the molecular level among four black goat breeds in Sichuan Province,and provide theoretical basis for rational protection and utilization.Method] The polymorphisms of mtDNA D-loop sequence and phylogenetic relationships among four black goat breeds in Sichuan were analyzed.Result] The fragment lengths of mtDNA D-loop in all black goat breeds were 1 210~1 212 bp,including five haplotypes,65 polymorphic sites,33 single polymorphic sites and 32 parsimony informative sites,and the average nucleotide diversity(π value)is 0.001 518.All black goat breeds have poor polymorphism in mtDNA D-loop sequence.Clustering analysis showed that Lezhi black goat and Jintang black goat cluster together at first,then both of them clustered with Jianchang black goat,and Zigong black goat cluster with all of above finally.Conclusion] All black goat breeds have poor polymorphism and low differentiation in mtDNA D-loop sequence.The clustering results agree well with zoology circumstance and geography distributing.
