| 摘 要： | 随着胚胎工程及胚胎相关技术的发展,绵羊卵母细胞和绵羊胚胎的需求量日益增加.获得尽可能多的结合型卵母细胞是体外受精的关键环节.本试验通过剖切法和抽吸法两种不同采卵方式获取卵母细胞并进行体外成熟培养.剖切法获得的卵母细胞数量高于抽吸法,所用时间相差不大.A、B级卵母细胞的比例在剖切法中显著高于抽吸法(P＜0.05),抽吸法所采集的C级卵母细胞显著高于剖切法(P＜0.05);剖切法中卵丘扩散率和第一极体排出率要显著高于抽吸法(P＜0.05),受精后不同时期胚胎的发育率差异不显著(P＞0.05).表明剖切法是采集绵羊卵母细胞的理想方法.

| Abstract: | With the development of embryonic project and relative embryonic technology,more and more sheep oocytes and embryos are requested.The key problem is how to obtain as many as combinative sheep oocytes.In this experiment,there were two different methods of harvesting oocytes in ovaries,aspiration and slicing,involved and its effect on subsequent maturation in vitro were studied.The results showed that the average total number of oocytes per ovary is significantly higher by slicing method than by aspiration method.but the time of recovery oocytes had no significant difference,and the percentage of grade C oocytes was significant higher by aspiration method than by slicing method(P<0.05).Besides,cumulus proliferate rate and primo polar body ejectment rate were significant higher by slicing method than by aspiration method(P<0.05).The developing rate of embryo in different period had no significant difference(P>0.05).In short,slicing method is an ideal method to get sheep occytes. |
