摘    要:本文利用热带种 (Badila)茎尖培养进行脱毒研究。试验结果表明 :细胞分裂素是茎尖成苗所必需的激素 ,萘乙酸则不是 ,0 .5- 1 .0 mm外植体在附加 2 .0 mg/ L BA和 0 .2 mg/ L NAA的 MS液体培养基中进行离体培养 ,成苗率可达 30 %以上 ;采用液体培养和在培养基中添加少量的活性炭均能有效地防止酚污染 ,提高成苗率 ;茎尖培养结合热处理能有效地脱除花叶病毒 ,侧芽培养无法进行脱毒。田间调查表明 ,茎尖脱毒苗在光合速率、株高、茎径方面显著优于未脱毒的侧芽苗 ,分蘖数两者相近

Abstract:With Badila as material, this paper studied on the methods of eliminating sugarcane mosaic virus (SCMV) by meristem tip culture. By means of liquid culture, 0.5-1.0 mm meristem tips were cultured in MS media supplemented with 2.0 mg/L BA and 0.2 mg/L NAA. The result indicated that the plantlet ratio was over 30%, and BA was necessary, but NAA not. Adding active carbon in liquid media was of benefit to preventing phenol pollution and increasing the plantlet ratio. It was shown that the method of meristem tip culture with heat treatment was effective in eliminating SCMV. However, it was impossible to eliminating SCMV by lateral bud culture. The results of observation in the field indicated that virus free plants derived from apical meristem were higher than infected plants derived from lateral bud in photosynthetic rate, stalk length and stalk diameter, but their tiller number was similar.
