湖北省恩施和远安稻区稻瘟病菌群体DNA的指纹分析及宗谱多样性检测

摘    要:从湖北恩施和远安2个稻区18个近等基因系上55个病斑中分离了113个稻瘟病菌的单孢菌株,并采用基于Pot2转座子序列的rep-PCR技术对该菌株群体进行了遗传多样性分析.结果表明:该菌株群体在DNA水平上具有丰富的多态性,以相似系数0.8为界,可将113个菌株划分为14个遗传宗谱;在不同近等基因系上,同一近等基因系上不同病斑中以及同一病斑中获得的菌株均可分属于不同的遗传宗谱;远安稻区的菌株均属于HB6和HB7,恩施稻区的菌株可分属于14个宗谱,提示恩施菌株的遗传多样性比远安菌株更丰富;属于同一宗谱的菌株可侵染不同近等基因系;2个优势宗谱HB6和HB7的菌株可分别侵染持有Pik、Pi3、Pi9(t)、Pi20、Pia、Pita 与Pii、Piz~5、Pib、Pi12、Pik~m的近等基因系.

Abstract:One hundred and thirteen single-spore rice blast pathogen isolates were obtained from 55 lesions on 18 near isogenic lines(NILs)in Enshi City and Yuan'an County in Hubei Province.Genetic diversity analysis of the population was conducted using rep-PCR amplification based on Pot2 transposon. Rich polymophisms were observed at DNA level and 113 isolates can be divided into 14 lineages at 0.80 level of genetic similarity coefficient.Isolates from different NILs,different lesions on the same NILs and the same lesion were classified into various lineages respectively.Isolates from Yuan'an county belonged to lineage of either HB6 or HB7.However,isolates from Enshi City can be distributed into 14 lineages,which implied the genetic diversity of isolates from Enshi City was richer than that of those from Yuan'an County.Moreover,isolates belonging to the same lineage were able to infect different NILs.Isolates from two dominant lineages,HB6 and HB7,were able to infect NILs carrying Pik,Pi3,Pi9(t),Pi20,Pia and Pita as well as Pii,Piz~5,Pib,Pil2 and Pik~m,respectively.
