保幼激素和蜕皮激素对家蚕离体后部丝腺吸收核酸和蛋白质标记前体的影响

摘    要:用后部丝腺离体培养和示踪原子法,研究了保幼激素类似物和植源性蜕皮激素对家蚕后部丝腺吸收核酸和蛋白质标记前体的影响。离体后部丝腺对~3H-胸腺嘧啶核苷、~3H-尿嘧啶和~3H-甘氨酸的吸收活性均受培养液中激素的影响。在本试验培养时间内,总吸收量均以两种激素同时加入高于单种激素加入;在加入其中一种激素时,吸收活性还因供试幼虫发育时期的不同(五龄前期或后期)而表现出相反的趋势;吸收的~3H-甘氨酸参入后部丝腺蛋白质合成的比率,以加入保幼激素类似物时较低,加入蜕皮激素和两种激素同时加入时较高。因此认为,不同发育时期接受过不同内源激素作用了的后部丝腺,在短期的体外条件下对激素的反应是不同的;但五龄任何时期的后部丝腺,均是在两种激素共同作用下,比单种激素作用更有利于核酸和蛋白质的合成。

Abstract:With the posterior silk gland culture in vitro and isotopic tracing,the effects of juvenile hormone analogues (JHA)and ecdysone(MH) on absorption of nuclear acid and protein marked precursors in the posterior silk gland was studied in this test. When the posterior silk gland was cultured in vitro, the absorptive activity of 3H-thymidine, 3H-uracil and 3H-glycocoll was all affected by hormones in culture media. The total absorption was higher when JHA and MH were given simultaneously than when JHA or MH was singularly given. The absorptive activity of the posterior silk gland appeared opposite trend due to different development stage of the larvae when only one hormone existed. The ratio of protein synthesis from 3H-glycocoll was lower when JHA was given than only MH or 2 hormones were given. These results suggested that the application of both JHA and MH at the same time in the 5th instar could be more advantageous to the synthesis of nuclear acid and protein than that of each hormone alone in the posterior silk gland.
