氨基酸对小麦成熟胚愈伤组织诱导率的影响

摘    要:为优化小麦成熟胚愈伤组织诱导体系,以中国春(Chinese Spring)成熟种子为材料,MS培养基为基础培养基,研究酒精、双氧水不同的灭菌时间和不同氨基酸组分对小麦成熟胚愈伤组织诱导与分化的影响。结果表明,当75%酒精浸泡时间控制在180 s、30%双氧水灭菌时间15 s以上时和当30%双氧水消毒时间控制在180 s、75%酒精浸泡时间15 s以上时,成熟胚均不会出现污染。甘氨酸、天冬氨酸、脯氨酸和亮氨酸是小麦成熟胚诱导的必须氨基酸。正交实验结果表明,培养基中最佳的氨基酸配组为甘氨酸60.0 mg·L-1、天冬氨酸2.0 mg·L-1、脯氨酸2.0 mg·L-1和亮氨酸0.1 mg·L-1。利用该培养体系,小麦成熟胚的出愈率达到97%以上。本研究优化了小麦成熟胚的培养体系,也为研究小麦氨基酸代谢的生理过程提供了参考。

Abstract:Optimization of transformation is essential in order to overcome the relatively low transformation rate encountered with wheat. We used the mature seed of China Spring and MS medium to study the effects of different amino acid medium and alcohol and hydrogen peroxide sterilization time on the callus induction. The results showed that the wheat embryo will not be contaminated when immersed in 75% alcohol for 180 s,and soaked in 30% hydrogen peroxide for longer than 15 s,or immersed in 30% hydrogen peroxide for 180 s,and soaked in 75% alcohol for 15 s or more. Glycine,aspartic acid,proline and leucine were essential amino acids in wheat mature embryo induction. The result of orthogonal design analysis on different compositions illuminated that the optimum medium was glycine 60.0 mg·L-1,aspartic acid 2.0 mg·L-1,proline 2.0 mg·L-1 and leucine 0.1 mg·L-1. And by this formula,callus rate of wheat matures embryo reached above 97%. This study not only further improves the tissue culture system of wheat mature embryo,but also provides a reference for studying the physiological process of amino acid metabolism in wheat.
