小麦温敏雄性不育系BNS366小孢子发育和花粉育性检测方法研究

摘    要:为了建立温敏雄性不育小麦BNS366小孢子发育和花粉育性的检测方法,以郑麦366作为对照,采用I2-KI法、醋酸洋红法、苏木精法和改良苯酚品红法染色观察小孢子发育过程,进行花粉可育率和自交结实率统计。结果表明,BNS366小孢子败育时期为单核靠边期至二核期。I2-KI法能清晰反映淀粉积累情况,但郑麦366在三核期和开花期看不到细胞核,其花粉可育率极显著高于自交结实率,操作最简便。醋酸洋红法能清晰显示细胞核,郑麦366花粉可育率与自交结实率一致,能够看到内容物的积累,但不清晰,操作简便。苏木精法在单核期和二核期染色效果最好,操作较复杂。改良苯酚品红法对细胞核和内容物的染色效果均不理想。因此,观察淀粉的积累情况以I2-KI法最好,观察细胞核的变化和检测花粉育性均以醋酸洋红法最好。

Abstract:In order to establish the method for detecting microspore development and pollen fertility in the thermos-sensitive male-sterile wheat line BNS366, with Zhengmai 366 as the control, the microspore development process was investigated by I2-KI method, acetate carmine method, haematoxylin method and carbol fuchsin method. Moreover, the pollen fertility and selfing seedset rates were measured at flowering stage. The results showed that the microspore abortion period of BNS366 was from the uninucleate period to the binucleate period. With the I2-KI method, the accumulation of starch in the microspore could be clearly reflected, but nucleus in microspore of Zhengmai 366 could not be detected at trinucleate period and flowering period. The pollen fertility rate of Zhengmai 366 was significantly higher than the selfing seedset rate at flowering period, and the operation was the easiest. With the acetate carmine method, the pollen fertility rate of Zhengmai 366 was consistent with the selfing seedset rate, and nucleus could be showed clearly and the operation was simple. The accumulation of contents in the microspores could be seen, but it was not clear. The haematoxylin method showed the best staining among the four methods at uninucleate period and binucleate period, but the operation was more complicated than that of I2-KI and acetate carmine method. With the carbol fuchsin method, the nucleus and contents could not be reflected clearly. Therefore, I2-KI method was the best for the accumulation of starch observation in the pollens. Moreover, acetate carmine method was the best for the changes observation of the nucleus and pollen fertility detection.
