| 摘 要： | 以传染性法氏囊病毒强毒株接种９日龄鸡胚尿囊腔，待鸡胚死亡后，收集其尿囊液，提取病毒。纯化的病毒ＲＮＡ在３％琼脂糖凝胶电泳上呈现出２９０ｂｐ和３４００ｂｐ的两条带。收集这两条带，以病毒全基因组制备生物素标记的探针。此探针有良好的特异性。它不与其它３种禽类病毒（ＮＤＶ，ＩＢＶ，ＩＬＶ）的核酸抽提物发生交叉反应。此试验是快速、敏感、可重复的。其敏感度达０．５ｐｇ。

Preparation of Biotin-labelled IBDV-RNA Probe and Its Specificity and Sensitivity in Detecting IBDV-RNA |

| Abstract: | Highly virulent strain of infectious bursal disease virus (IBDV) was propagated inallantois of 9-day old embryonated eggs.While the embrycs died, the allantoic fluid was harvestedand the IBDV was recovered. Purified dsRNA from IBDV showed on 3%agarose gel two bandswhich were 3400bp and 2900bp, respectively.A biotin-labelled probe was prepared using the wholegenome of IBDV. Preliminary test revealed that this probe was highly specific. It did not hybridizeto the nucleic acids of other three poultry viruses (NDV, IBV and ILV). Its sensitivity reached0.5pg. |
