茭白黑粉菌耐热突变体的辐射诱变选育与生长特征研究

摘    要:来自茭白正常膨大肉质茎的黑粉菌(M—T菌株)经5kGy和10kGy 60Co-γ射线辐照后,筛选获得耐40℃的耐热突变体(H—T菌株)各10个菌株,而来自灰茭的黑粉菌(T菌株)未观察到耐热突变体。对3种菌株在不同温度下的生长特性作了初步分析。研究结果表明,M—T和T菌株均不能在32℃及以上温度生长;15~28℃范围内,H—T菌株的生长速率显著快于T菌株,后者又显著快于M—T菌株;相对于最适温度下的延滞期,显著延长延滞期的临界温度分别为15℃(M-T菌株)、20℃(T菌株)和20℃(H-T菌株)。因此,H-T菌株与M-T菌株的生长特性存在较大差异。

Abstract:Ustilago esculenta mutants (H-T strain), tolerant of 40~C, were obtained from M-T strain irradiated with 5 kGy and 10 kGy of 60Co-γ ray, but no thermo-tolerant mutant was obtained in T strain from U. eseulenta. And then we studied the growth characteristics of M-T, T and H-T strains at 15-40℃. The results showed that both M-T and T strains were unable to grow while the temperature was above 32℃, and H-T strain grew significantly quicker than T strain at 15-28℃, but T strain grew significantly quicker than M-T strain. The critical temperature significantly prolonged lag phase was 15℃ for M-T strain, 20℃ for T strain and 20℃ for H-T strain respectively, compared to those at optimal temperature. In sum, the growth characteristics of the thermo-tolerant H-T strain had large differences with those of M-T strain.
