摘    要:以猪红细胞膜抗原免疫BALB/c小鼠,应用杂交瘤技术将免疫小鼠的脾细胞与Sp2/0骨髓瘤细胞融合,通过微孔板凝集试验及玻片凝集试验筛选可稳定分泌非凝集型mAb的杂交瘤细胞株,并用有限稀释法进行克隆.制备出一株可稳定分泌非凝集型抗体的杂交瘤细胞株1C2,其亚型为IgG1类,培养上清和腹水效价分别为1:1024和1:108,没有种属交叉血凝反应,为构建双特异性抗体奠定了基础.

Abstract:BALB/c mice were immunized with the membrane antigens of swine erythrocytes.The splenocytes of the immunized mice were fused with Sp2/0 myeloma cells by hybridoma technique.The antibodies to common antigen of swine erythrocytes were screened by micro-quantity plate agglutination test method and then the agglutinating antibodies were weeded out by slide hem-agglutination assay.The hybridoma cells secreting non-agglutinating antibodies were cloned by end dilution method.One hybridoma cell 1C2 was obtained, which secreted non-agglutinating antibody.The mAb 1C2 belonged to IgG1, and without specie scross-agglutination reaction.The titers of cultures suppernatant and ascites of 1C2 were 1:1024 and 1:108 respectively.The mAb 1C2 has high titer, affinity and specificity, which could be used for the preparation of bi-specific antibodies (BsAb).
