苦瓜子蛋白分离纯化的研究

摘    要:把苦瓜子粉碎后用50 mmol/L乙酸抽提、硫酸铵沉淀,经透析得到蛋白粗提液,然后用考玛斯亮蓝G-250法测蛋白质含量为16.517μg/g;SDS-PAGE分析表明,苦瓜种子含有3种蛋白组分,相对分子质量分别为29.5,22.0和9.6 kDa,其中分子量为29.5 kDa的含量最多,9.6 kDa的含量次之,22.0 kDa的含量最少。再对粗提液过Sephadex G-100凝胶柱进一步纯化得到4个主峰,吸光度分别为0.62,0.28,0.36,0.42;经SDS-PAGE检测,得知第1,2峰分子量均为29.5 kDa。

Abstract:Seeds suffer the crush,50 mmol/L acetic acid and extraction, ammonium sulfate precipitation, the crude protein by dialysis fluid and then test Thomas-blue G-250 was measured and the results showed that the crude protein content was 16.517 μg/g, SDS-PAGE analysis showed that bitter gourd seeds containing three protein components, the molecular weights were 29.5,22.0 and 9.6 kDa,which molecular weight of 29.5 kDa was the most,9.6 kDa the second,22 kDa the least. To the crude extract of a Sephadex G-100 gel column further purified by the four main peaks,absorbances were 0.62,0.28,0.36 and 0.42.The SDS-PAGE detection showed that the first and second peaks were all 29.5 kDa.
