应用MABA法与试管法测定蛇床子素和8-甲氧基补骨脂素体外抗结核活性的研究

摘    要:为筛选有开发价值的抗结核中草药,本研究通过MABA(Microdilution MABA Assay)与试管法分别测定蛇床子素和8-甲氧基补骨脂素对结核分支杆菌H37Rv(ATCC27294)和H37Ra(ATCC25177)的体外抗菌活性,即最低抑菌浓度(MIC),同时比较两种方法的检测结果;并以MTT法研究蛇床子素和8-甲氧基补骨脂素对Balb/C小鼠的脾细胞和肝细胞的细胞毒性试验。蛇床子素和8-甲氧基补骨脂素对这两种结核菌的最低抑菌浓度分别均为32μg/mL和128μg/mL,并且对BALB/c小鼠的脾细胞和肝细胞均无毒性。MABA法与试管法相比较,完全符合率为90%(9/11),具有较好的一致性。本研究为蛇床子素和8-甲氧基补骨脂素作为抗结核药物的开发及它们的抗菌机制研究奠定了基础。

Abstract:The antimycobacterial activities of Osthole and 8-MOP against Mycobacterium tuberculosis H37Rv (ATCC 27294) and H37Ra (ATCC 25177) were determined by the microdilution MABA Assay (MABA) and the conventional two folds dilutions method for screening candidate herbs. The cytotoxicity was determined by MTT method in splenic cell and hepatic cells of BALB/c mouse. Osthole showed a minimal inhibitory concentration (MIC) of 32 μg/mL against M. tuberculosis H37Rv and H37Ra, and 8-MOP showed a MIC of 128 μg/mL against M. tuberculosis H37Rv and H37Ra. Osthole and 8-MOP were no cyto-toxicity to the cells. The results would pave the way to further development and exploring the mechanism of Osthole and 8-MOP against Mycobacterimn.
