不同游离方式辣椒小孢子的胚胎发生

摘    要:以甜椒、牛角椒、羊角椒、线椒和朝天椒5个不同类型的10个辣椒品种为试材,研究通过直接机械游离、看护培养自然游离和先花药预培养再机械游离3种不同的小孢子游离方式的胚胎发生情况。结果表明,先花药预培养再机械游离小孢子的胚胎发生最好,看护培养自然游离小孢子的胚胎发生次之,直接机械游离小孢子的胚胎发生最差,仅获得少量球形胚;小孢子在培养前期需要花药壁的存在,高温预处理之后则需要花药壁及时破裂。

Abstract:Embryogenesis of microspores through three different isolation methods,including direct mechanical isolating,shed-culture nature isolating,and mechanical isolating after anther pre-culture,were studied by using 10 Capsicum varieties belonging to 5 different fruit types of pepper as the materials.The results showed that embryogenesis of microspores through mechanical isolating after anther pre-culture was the best,then shed-culture nature isolating,and direct mechanical isolating was the worst,which got only some global embryoids;anther wall was necessary to microspores at the beginning of culture,and must be ruptured in time after high temperature pre-treatment.
