摘    要:目的]优化鸡腿菇子实体多糖的提取工艺。方法]以水为浸提液,通过单因素试验研究了浸提温度(60、70、80、90℃)、浸提时间(2、3、4、5h)、料液比(1:10、1:20、1:30、1:4Jo)对鸡腿菇子实体多糖得率的影响,并采用正交试验对提取工艺进行优化。结果]苹因素试验表明,最佳浸提温度、浸提时间和料液比分别确定为4h、90℃、1:30。正交试验表明,浸提温度对鸡腿菇多糖得率的影响最太,其次是浸提时间,液固比影响最小。通过对提取条件的优化,结合收益、成本等综合因素选出适合本地条件的优化工艺为:浸提温度90℃、浸提时间3h、料液比1:30。验证试验显示,在最佳工艺条件下提取的多糖得率达7.99%。结论]该优化工艺的回收率高迭98%,说明工艺条件较稳定,适用于工业化生产。

Abstract:Objective] The aim was to optimize the extraction technology of polysaccharides from Coprinus comatus fruit-body. Method] With the water as the extraction solvent, the effects of extraction temperature (60, 70, 80, 90 ℃), extraction time (2, 3, 4, 5 h), solid-liquid ratio (1∶ 10, 1∶ 20, 1∶ 30, 1∶ 40) on the yield of polysaccharides from C. comatus fruit-body were investigated by the single factor experiments and the extraction technology was optimized by using the orthogonal experiment. Result] The single factor experiments showed that the optimum extraction temperature, extraction time and solid-liquid radio were 4 h, 90 ℃, 1∶ 30 resp. The orthogonal experiment showed that the effect of extraction temperature on the yield of polysaccharides from C. comatus was the largest, the second was the extraction time and the effect of solid-liquid radio was the least. The optimization technology suitable for local conditions which was selected through optimizing extraction condition and combining the comprehensive factors of benefit, cost and so on was as follows: the extraction temperature was 90 ℃, the extraction time was 3 h and the solid-liquid radio was 1∶ 30. The verification test showed that the yield of polysaccharides extracted under the optimum technology condition was 7.99%.Conclusion] The recovery rate of the optimization technology was up to 98%, showing that the technology condition was stable and suitable for the industrial production.
