摘    要:为了确定不同来源的平菇遗传差异,利用相关序列扩增多态性(sequence-related amplified polymorphism,SRAP)分子标记技术,对安徽地区的16个平菇品种的遗传多样性进行了分析。筛选出24对随机引物组合,获得了627条清晰条带,平均每对引物产生26.1条。多态性条带共189条,占总条带数的30.1%,平均每对引物组合得到7.88条多态性条带。依据SRAP标记聚类,在遗传相似系数0.65的水平上,供试的16个平菇品种分为2个大类:Ⅰ和Ⅱ。Ⅰ类分为2个亚类:A类和B类,Ⅱ类也分为2个亚类:C类和D类。其中A类和D类各包括4个品种,B类中有3个品种,C类中有5个品种,从而在DNA水平上证明了所研究的平菇种质资源的遗传多样性。

Abstract:In order to assess the genetic diversity ofPleurotus ostreatus from different sources,sequence-re lated amplified polymorphism (SRAP) molecular markers were used to detect the genetic diversity of 16 species ofPleurotus ostreatus in Anhui.A total of 627 amplified fragments and 189 polymorphic fragments were detected using 24 pairs of random primer combinations.The polymorphic fragment percentage was 30.1%.The number of amplified fragments and polymorphic fragments per primer combination were 26.1 and 7.88,respectively.Sixteen species of Pleurotus ostreatus materials were divided into Ⅰ and Ⅱ at genetic distance of 0.65.Group Ⅰ could be divided into A and B sub-groups,and group Ⅱ could also be divided into C and D sub-groups.There were four species in A and D subgroups,3 in B subgroup and 5 in C subgroup.Thus the genetic diversity of the Pleurotus ostreatus varieties was confirmed at the DNA level.
