山东小麦黄花叶病毒的分子检测与鉴定

摘    要:利用RT-PCR对2012年山东10个地区送检的105个小麦样品进行了检测,从泰安、临沂、枣庄3个地区的样品中检测到了小麦黄花叶病毒(Wheat yellow mosaic virus,WYMV)。这3个地区WYMV分离物CP基因的核苷酸一致率为97.6%~98.3%,氨基酸一致率为98.3%~99.0%。在利用CP基因核苷酸序列构建的系统进化树中,这3个分离物均与WYMV分离物聚类到一起,其中枣庄分离物和河南分离物(FR828007)亲缘关系更近。

Abstract:One hundred and five wheat samples collected from 10 districts of Shandong Province were detected for wheat yellow mosaic virus(WYMV) using RT-PCR.WYMV was detected from samples from Taian,Linyi and Zaozhuang.The coat protein genes of WYMV from these three areas shared the nucleotide identity of 97.6%~98.3% and the amino acid identity of 98.3%~99.0%.In the phylogenetic tree constructed based on the nucleotide sequence,the three isolates were clustered together with other WYMV isolates.The isolates from Zaozhuang had closer genetic relationships with the isolate from Henan.
