茶树ACC合成酶基因全长cDNA的克隆及其生物信息学分析

摘    要:乙烯是一种重要的植物激素,参与植物许多生理过程。ACC(1-氨基环丙烷-1-羧酸)合成酶(ACS)是植物乙烯合成过程中的限速酶,对乙烯的合成具有重要的调控作用。利用其它植物ACS基因的保守序列设计兼并引物,采用RACE(Rapid Amplificationc DNA Ends)和RT-PCR等技术,克隆得到编码茶树ACS基因的全长cDNA序列,长1579bp,编码478个氨基酸,预测分子量约为53.7kD,等电点8.039,GenBank登录为EF205149。以Neighbor-Joining法构建进化树,发现茶树ACS基因与柿树中的同类基因亲缘关系最近。茶树ACS基因的克隆为从分子水平认识乙烯在茶树上的生理作用奠定了基础。

Abstract:Ethylene is one of the important plant hormones. It has significant physiological role in the life of plant. ACC (1-aminocyclopropane-1-carboxylic acid) synthase (ACS) is a key enzyme in the ethylene biosynthesis in plants. It plays important roles in the regulating ethylene biosynthesis. Degenerate primers were designed based on the conservative ACS gene sequence of other plants in the GenBank to clone the full-length cDNA of ACS genes of the tea plant by using the RACE and RT-PCR methods. It is deposited to the GenBank (Accession No. EF205149). The tea plant ACS gene cDNA is 1 579 bp in length, encoding 478 amino acids, the putative molecular weight is 53.7 kD and the pI is 8.039. The phylogenetic trees constructed by the software MEGA 3 showed that ACS gene of tea plant was closely related to that of Diospyros kaki. The cloning of ACS gene full-length cDNA from the tea plant provides a possibility for understanding the molecular mechanism of physiological role on molecular level in the tea plant.
