急性热应激对麻鸡免疫功能的影响

摘    要:研究急性热应激对麻鸡免疫机能的影响,将30只28日龄麻鸡随机分成5组,即对照组、热应激3 h组、热应激6 h组、热应激9 h组和热应激12 h组,每组6只。将麻鸡在37℃±1℃条件下分别处理3、6、9、12 h,对照组麻鸡于室温条件下常规饲养。应激试验后取脾脏和法氏囊制成石蜡切片,分别进行HE染色和γ干扰素(IFN-γ)的免疫组化检测,并通过蛋白免疫印迹的方法检测麻鸡脾脏中HSP70、IFN-α、IFN-β、IFN-γ、NF-κB p65的蛋白含量水平变化。37℃热应激条件下3 h^12 h麻鸡脾脏无明显的组织学变化;法氏囊中的淋巴滤泡从热应激3 h后开始增大,淋巴滤泡之间排列更加紧密,淋巴细胞增多;脾脏中IFN-α和IFN-β在热应激9 h后显著升高(P<0.05),IFN-γ在6 h时显著升高(P<0.05),9 h后恢复正常值。HSP70在应激3 h出现短暂下降后持续升高,而NF-κB p65在热应激3 h时出现短暂的波动后持续下降,显著低于对照组(P<0.05)。结果表明,37℃条件下热应激12 h内能够提高麻鸡机体免疫能力,有利于麻鸡生长发育。

Abstract:To study the effect of heat stress on immune function of Ma chickens at different time,30 Ma chickens(twenty-eight-day-old)were randomly divided into 5 groups:control group,acute heat stress 3 h group,heat stress 6 h group,heat stress 9 h group and heat stress 12 h group.Chickens were treated at 37℃for 3,6,9 and 12 h respectively.The control group chickens were kept at room temperature,and all the chickens were given with normal drinking water.At the end of the experiment,the spleen and bursa of Fabricius were prepared into paraffin sections for H&E staining and immunohistochemistry for gamma interferon(IFN-γ),and spleen protein levels of HSP70,interferon-α(IFN-α),interferon-β(IFN-β),interferon-γ(IFN-γ)and NF-κB p65 were detected by Western blot.The results showed that there were no obvious histological changes in the spleen under 37°C heat stress within 12 h,the lymphocytes in the bursa of Fabricius increased from 3 h after heat stress,and the lymphoid follicles were more closely arranged and lymphocytes increased.IFN-αand IFN-βincreased significantly after heat stress for 9 h(P<0.05),and IFN-γreturned to normal at 9 h after a significant increase at 6 h(P<0.05).HSP70 continued to increase after a brief decrease in stress for 3 h,while NF-κB p65 continued to decrease after a brief fluctuation in heat stress for 3 h,which was significantly lower than that of the control group(P<0.05).The results showed that 37℃heat stress within 12 h can significantly improve the immunity of the Ma chickens,which is conducive to growth and development.
