| 摘 要： | 为建立一种快速鉴别猪流行性腹泻病毒(PEDV)G1型/G2型感染的RT-PCR方法,基于PEDV的S基因和E基因序列分别设计了PEDV的分型引物和鉴别引物,经过敏感性试验、重复性试验等一系列反应条件的优化,成功建立了鉴别PEDV G1/G2型感染的双重RT-PCR方法;通过目的片段的数量来判断PEDV的感染类型,其中,PEDV G1型感染可扩增得到一条目的片段(249 bp),G2型感染扩增得到两条目的片段(840 bp和249 bp)。利用该方法对采自广西不同地区的92份临床样品进行检测,结果显示,阳性样品55份,其中,G2型样品53份,G1型样品2份。结果表明该方法操作简易,具有良好的敏感性和特异性,能快速鉴别PEDV G1/G2型的感染。

| Abstract: | In order to establish a rapid RT-PCR assay for differentiation of G1/G2 of porcine epidemic diarrhea virus(PEDV),the typing and detected primers were designed respectively based on the S and E gene sequences of PEDV.Through experiments of sensitivity,specificity and repeatability,a duplex RT-PCR were established successfully for identification of G1/G2 infections by amount of fragments.G1 strain of PEDV generated a fragment(249 bp),and G2 strain produced two fragments(840 bp and 249 bp).92 clinical specimens collected from different farms in Guangxi were detected by the established RT-PCR.The results indicated that 55 samples were positive for PEDV,of which 53 were positive for G2,2 for G1.It suggests that the method can be used to identify G1/G2 rapidly with simple operation,great sensitivity and specificity. |
