新型标记SRAP及其在草业研究中的应用前景

摘    要:相关序列扩增多态性(Sequence-related Amplified Polymorphism,SRAP)是最近发展起来的新型分子标记,具有简便、可靠、易于测序等优点。SRAP独特的引物设计使其可检测基因的可阅读框(ORFs)区域,正、反向引物分别与外显子和内含子(或启动子)区域配对,因不同物种、不同个体的内含子、启动子与间隔区长度不等而产生多态性,目前已开发出多个SRAP正、反向引物;SRAP-PCR扩增程序采用复性变温法,即前5个循环复性温度为35℃,后35个循环为50℃;目前SRAP已在植物图谱构建、遗传多样性评价、QTL定位以及杂种优势预测等方面成功应用。在草业研究方面,已有将SRAP标记用于野牛草种质资源遗传多样性分析的报道,其在草业研究中的应用前景十分广阔。

Abstract:Sequence-related amplified polymorphism(SRAP)is a newly developed molecular marker with the advantages of simplicity,reliability and ready to sequence.It is based on two-primer amplification and aimed for amplification of open reading frames(ORFs)by special primer set.The forward primer amplified exons,the reverse primer amplified introns(or prompters).The polymorphism fundamentally originates from the variation of the length of these introns,promoters and spacers,both among individuals and among species.Some forward primers and reverse primers were developed recently.SRAP-PCR amplified procedure change the annealing temperature,i.e.the first five cycles the annealing temperature is set at 35°C,and the following 35 cycles are run at 50°C.This technique are being applied in genetics mapping,genetics diversity analysis,QTL tagging and heterosis prediction.There were genetics diversity analysis of buffalograss and the prospect of the SRAP marker application in pratacultural research is wide.
