摘    要:目的]探索左氧氟沙星的测定方法。方法]以磷酸二氢钾-磷酸-三乙胺/乙腈(851:5)为流动相,在1 000 ml磷酸缓冲液中含6.5g磷酸氢二钾、4ml磷酸、5ml三乙胺,流速1ml/min,柱温40℃、检测波长295 nm和pH值为3.5的条件下,通过反相HPLC测定兔血浆中左氧氟沙星的浓度,并分析其回收率。结果]通过该方法将左氧氟沙星与血浆成分较好地分离。左氧氟沙星的线性范围为0.005~50μg/ml,平均回收率为98.40%左右,最小检测浓度为50 ng/ml。结论]该方法具有操作简便、回收率高和重现性好等优点,可快速测定人血液中左氧氟沙星的浓度和检测禽畜体内的左氧氟沙星残留。

Abstract:Objective] The aim of the research was to discuss the determination method of levofloxacin.Method] With KH2PO4-H3PO4-triethylamine/acetonitrile(85:15) as mobile phase,under these conditions as adding 6.5 g KH2PO4,4 ml H3PO4 and 5 ml triethylamine in 1 000 ml phosphate buffer,flow velocity of 1 ml/min,the column temperature of 40 ℃,the detection wavelength of 295 nm and pH value of 3.5,the concn.of levofloxacin in rabbit plasma was determined by reversed-phase HPLC and its recovery was analyzed.Result] Levofloxacin and plasma components were separated better by using this method.The linear range of levofloxacin was 0.005-50 μg/ml,the average recovery was about 98.40% and the minimum detection concn.was 50 ng/ml.Conclusion] This method had such advantages as simple operation,high recovery and good repeatability.It could determine the concn.of levofloxacin in human blood quickly and detect the residue of levofloxacin in animal and poultry bodies.
