摘 要: | 目的]提高草莓花药培养效率。方法]以草莓品种“丰香”为材料,研究了不同条件对诱导花药愈伤组织和绿苗分化的影响。结果]8℃预处理3d,草莓花药的愈伤组织诱导率可达64.8%,显著高于处理5d(46.5%)和不经低温处理(40.3%)的诱导率。22℃和25℃培养时,愈伤组织诱导率分别为61.30%和68.36%,前者的愈伤组织优于后者。激素组合BA 0.2mg/L+KT 1.0mg/L+NAA 2.0mg/L的出愈率达61.57%,且愈伤质量较好。黄绿色颗粒状、绿色致密、淡黄色蓬松等3种愈伤组织分别占11.5%、49.0%、39.5%。愈伤组织在激素组合ZT 2.0mg/L+BA 0.5mg/L+NAA 0.2mg/L中的分化率最高,可达11.28%。生根培养后,花培植株生根率达98%。结论]得到了草莓花药培养的适宜条件。

Abstract: | Objective] The study was to improve the culture efficiency of strawberry anther.Method] With strawberry variety Fengxiang as the tested material,the effect of different condition on the induction of anther callus and differentiation of green plantlet was investigated.Result] The induction rate of strawberry anther callus in the pretreatment for 3 d at 8 ℃ could reached 64.8%,being obviously higher than that in treatment for 5 d(46.5%) and that in the treatment without low temperature(40.3%).When the cultured was at 22 ℃ and 25 ℃,the induction rate of callus was 61.3% and 68.36% resp.and the callus of the former was better than that of the latter.The callus rate from the hormone combination of BA 0.2 mg/L + KT 1.0 mg/L +NAA 2.0 mg/L was 61.57% with a better quality. 3 kinds of callus such as the yellow-green granular,green and compact,slight yellow fluffy accounted for 11.5%,49.0% and 39.5% resp.The differentiation rate of callus from hormone combination of ZT 2.0 mg/L +BA 0.5 mg/L +NAA 0.2 mg/L was the highest,being 11.28%.After rooting culture,the rooting rate of anther culture plant was 98%.Conclusion] The optimum condition for the strawberry anther culture was obtained. |
