棉花枯萎病菌激活蛋白的提取及其对棉花种子发芽的影响

摘    要:为探索棉花枯萎病的生物防治方法,对棉花枯萎病菌激活蛋白的提取及活性进行了初步研究。首先从感染枯萎病的棉花组织中分离培养棉花枯萎病菌,置于不同温度下以不同转速进行培养,获得该菌的最佳发酵条件为26℃、200r/min、72h,菌丝干质量可达3.611g,为大量提取棉花枯萎病菌激活蛋白奠定了基础。对棉花枯萎病菌的激活蛋白进行提取后,电泳检测发现,在分子量为51kD附近有一条较为集中的电泳谱带。用激活蛋白1 000倍、1 500倍、2 000倍稀释液分别处理2个棉花品种(新陆中7号、新陆中32号)的种子,以清水作对照,检测激活蛋白对棉花种子发芽的影响。结果表明,激活蛋白能够提高2种棉花种子的发芽率、促进胚根生长,其中稀释1 500倍时的促进效果最明显,分别为:新陆中7号发芽率90%,胚根长6.04cm,胚根鲜质量85.78mg,胚根干质量8.89mg;新陆中32号发芽率85%,胚根长5.12cm,胚根鲜质量77.41mg,胚根干质量8.56mg。表明棉花枯萎病菌激活蛋白具有高效的诱导激活活性,为研制低成本的蛋白生物农药提供了理论依据。

Abstract:In order to explore biological control technologies of cotton Fusarium wilt,the extraction method and activity of activator proteins from Fusarium oxysporum were studied preliminarily in this article.F.oxysporum was isolated from cotton tissues infected by Fusarium wilt.The best culture conditions,26 ℃,200 r/min and 72 h were obtained by culturing F.oxysporum at different temperatures,speed and time.And the dry weight of mycelia was up to 3.611 g under this condition.The extracted proteins exhibited a relatively centralized electrophoretic band with the molecular weight of about 51 kD.The cotton seeds of two varieties(Xinluzhong No.7,Xinluzhong No.32) were treated by the activator protein which was diluted 1 000 times,1 500 times and 2 000 times,and sterile water was used as CK.The effect of the activator protein diluent on seed germination was detected,and the results indicated that it could promote seed germination and radicle growth of the two cotton seeds,with 1 500-time diluent performing remarkable effects.For Xinluzhong No.7,the seed germination rate was 90%,the radicle length was 6.04 cm,and the fresh and dry weight of radicle was 85.78 mg and 8.89 mg.The results were 85%,5.12 cm,77.41 mg and 8.56 mg,respectively,for Xinluzhong No.32.These indicated that the activator protein had efficient induction activity,which could provide theoretic basis for development of low-cost protein biological pesticides.
