麒麟菜膳食纤维对SD大鼠30天喂养实验和致畸作用

摘    要:为评价麒麟菜(Eucheuma)膳食纤维的食用安全性,按照中国食品安全性毒理学评价方法对SD大鼠进行了30d喂养和传统致畸实验。30d喂养实验,分对照组和低、中、高剂量组,分别将麒麟菜膳食纤维用0%、1%、5%、10%的剂量添加于饲料中通过30d混饲方式给予受试物,实验期间观察其一般状况、体重、摄食量、食物利用率。第30天处死各组大鼠,对血液学指标、血清生化指标、器官以及组织病理学检查。传统致畸实验,分阴性对照组和10%饲料添加量的剂量组,另设阳性对照组,以浓缩鱼肝油25mL/kgBW灌胃,实验期间按时检查孕鼠体重,实验末期取胎检查其外观、记录死、活胎数及骨骼和内脏的异常情况。结果表明,30d喂养实验中,动物生长正常,无一死亡或中毒症状,各项观察指标不随剂量的增大和受试时问的延长出现剂量反应关系;致畸实验中,孕鼠生长良好,胚胎发育正常,胎鼠异常率均在自然发生率范围内。因此,可认为麒麟菜膳食纤维无毒、无致畸作用,食用安全可靠。

Abstract:In order to assess the food safety of dietary fibre(DF) from Eucheuma,a 30 d feeding experiment and a teratogenicity analysis were conducted in accordance with the methods of China for toxicological assessment on food safety.The thirty-day-feeding experiment was carried out by administering medicated feed with different doses at 0%,1%,5% and 10% of DF in diets corresponding with control,low-dose,mid-dose and high-dose groups.During the test,the general condition,body weight,feed consumption and feed conversion efficiency were examined.After thirty days feeding,the rats were killed and hematology parameters,serum biochemistry parameters,organs and histopathology were examined.Teratogenicity test was carried out by administering medicated feed at doses of 0% and 10% corresponding with negative control and dose-group;another positive control was designed by administering fish liver oil at dose of 25?mL/kg BW.During the test,the body weight of pregnant rats was examined on time,and at the end of the test,the pregnant rats were killed and the facies of fetuses,living and dead fetuses,ratio of abnormal bone and abnormal harslet were examined.The results showed that all the rats grew constantly,no death nor toxic symptom.All the observed index showed no relationship between postponing time and increasing doses during 30?d feeding period.And in teratogenicity tests,pregnant rats grew well;fetuses developed naturally,and the ratio of abnormal fetues was in the range of natural abnormal ratio.It is concluded that dietary fibre from Eucheuma was edible and safe as food without toxicity and teratogenic effect.Journal of Fishery Sciences of China,2006,13(1):65-72]
