巴西人参组培快繁体系的建立

摘    要:以巴西人参带芽茎段为外植体,研究培养基、植物生长调节剂、移栽基质等对巴西人参组培快繁的影响。结果表明:最佳的初代诱导培养基为WPM+6-BA 0.4 mg/L+NAA 0.4 mg/L,初代芽诱导率达90.65%,每个外植体平均萌芽数为2.90个,平均芽高2.70 cm;最佳的继代增殖培养基为WPM+6-BA 2.0 mg/L+NAA 0.2 mg/L,采用平铺的方式接种,培养25 d增殖系数可达5.21;最佳的生根培养基为1/2WPM+6-BA 0.3 mg/L+NAA 0.5 mg/L+PP333200,培养9.85 d即可生根,生根率高达98.85%,株平均生根数为18.87条;选择椰糠︰谷壳=4︰1(v/v)作为移栽基质,移栽成活率最高,可达98.67%。本研究建立的组培快繁技术体系可适用于巴西人参工厂化育苗生产,为巴西人参种苗快速繁殖提供技术保障。

Abstract:Using stem with buds as explants,the effects of culture medium,plant growth regulator and transplanting matrix on the tissue culture of Pfaffia paniculate was studied.The results showed that the optimal medium for initial induction was WPM+6-BA 0.4 mg/L+NAA 0.4 mg/L,the induction rate of initial buds was 90.65%,the average number of buds was 2.90 and the average height of buds was 2.70 cm.The optimal subculture culture medium was WPM+6-BA 2.0 mg/L+NAA 0.2 mg/L,and the proliferation coefficient of 25 d was 5.21.The best rooting medium was 1/2WPM+6-BA 0.3 mg/L+NAA 0.5 mg/L+PP333200 mg/L,the average time of root formation was 9.85 days,the rooting rate of buds was 98.85%,and the average number of rooting was 18.87.Coconut bran︰chaff=4︰1(v/v)was selected as transplanting substrate,the transplanting survival rate was the highest,up to 98.67%.The technology system of tissue culture and rapid propagation established can be applied to the industrial seedling production of P.paniculate,which provides technical guarantee for rapid propagation of P.paniculate seedlings.
