产类胡萝卜素的海洋红酵母的培养条件优化研究

摘    要:目的]优化产类胡萝卜素的海洋红酵母的培养条件。方法]以海洋红酵母Z44为试验菌株对其培养条件进行优化,同时,研究了添加物及培养条件对细胞量及类胡萝卜素产量的影响。结果]得到的培养基组成为:蔗糖25.0 g/L,酵母膏20.8 g/L,草酸铵4.2g/L,KH2PO44.0 g/L,Na2HPO44.0 g/L,MgSO4.7H2O 0.2 g/L,CaCl20.2 g/L,氟化铵10 mg/L,柠檬酸30 mg/L,L-亮氨酸30 mg/L,L-缬氨酸40 mg/L。在培养基中加入氟化铵、柠檬酸、L-亮氨酸及L-缬氨酸时,红酵母的类胡萝卜素产量是对照值的1.5倍。对培养条件的优化结果为:装液量为250 ml三角瓶中装60 ml的培养基,接种量8%,培养温度30℃,初始pH为5.5,培养结束时间为66 h,培养条件优化后,红酵母的生物量及类胡萝卜素产量分别为28.2 g/L和6.72 mg/L,分别是未优化时的1.26倍和1.87倍。结论]研究可为海洋红酵母生产类胡萝卜素的工业化应用提供参考。

Optimization of Fermentation Conditions for Carotenoids Production by Rhodotorula sp

Abstract: Objective] To optimize fermentation conditions for production of carotenoids by Rhodotorula sp. Method] The culture conditions for the production of carotenoids by Rhodotorula sp. Z44 strain were optimized. At the same time, effects of additives and culture conditions on cell biomass and carotenoid yield were studied. Result ] The optimum medium contained (g/l) 25.0 sucrose, 20.8 yeast extract, 4.2 ( NH4 ) 2 C2 04, 4.0 KH2 PO4, 4.0 Na2 HPO4, 0.2 MgSO4, 0.2 CaC12, (mg/1) 10 NH4 F, and 30 C6 H8 07, 30 C6 H13 NO2, 40 C5 H11 NO2. When ammonium fluoride, citric acid, L-leucine and L-valine was added to culture medium, Rhodotorula sp. carotenoids yield was 1.5 times as the control value. In a shake flask, the optimal culture conditions were as follows : initial pH 5.5, medium volume 60 ml ( in 250 ml flask), Inoculum size 8% and culture time 66 h at 30 ℃. Under optimum conditions, Rhodotorula sp. biomass and carotenoids production reached 28.2 g/L and 6.72 mg/L respectively, which were respectively 1.26 times and 1.87 times as those of the original medium. Conclu- sion] The study can provide reference for industrialization application of Rhodotorula sp. Carotenoids.
