油茶根际土壤解钾菌的发酵培养

摘    要:采用双层平板法对已分离出的15株油茶根际解钾细菌进行筛选,获得3株解钾能力较强的菌株CoPDB 6、CoPDB 13和CoPDB 15,并对3个菌株的培养基、pH、装液量、接种量、温度和摇床转速等发酵培养条件进行优化。结果表明,解钾细菌的最佳发酵培养条件为培养基A(蔗糖5.0 g,磷酸氢二钠0.5 g,云母粉1.0 g,MgSO4·7H2O 0.5 g,氢化铁0.005 g,水1000 mL)、培养温度30℃、pH值7.2~7.4、装液量20%~30%(50~75 mL/250 mL)、接种量9%、摇床转速200 r·min-1

Abstract:The 15 isolated potassium-dissolving bacteria in the rhizosphere of Camellia oleifera were screened by the double-layer plate method,three strains CoPDB 6,CoPDB 13 and CoPDB 15 with strong potassium-dissolving ability were obtained.The fermentation and culture conditions of the three strains such as culture medium,pH,liquid volume,inoculum volume,temperature and shaker speed were optimized.The results show that the best medium for potassium dissolving bacteria is medium A(Sucrose 5.0 g,disodium hydrogen phosphate 0.5 g,mica powder 1.0 g,MgSO4·7H2O 0.5 g,ferric chloride 0.005 g,water 1000 mL),and the most suitable fermentation culture condition is culture the temperature is 30℃,the pH value is 7.2~7.4,the liquid volume is 20%~30%(50~75 mL/250 mL),the inoculation volume is 9%,and the rotating speed of the shaker is 200 r·min-1.
