土壤颉颃芽孢杆菌B-Q2-7的分离筛选及其抑菌作用研究

摘    要:从杨凌大寨乡采来的土壤中分离到142株芽孢杆菌,经过皿内对峙初筛和无菌发酵滤液复筛得到一株对稻瘟病菌,小麦赤霉病菌,黄瓜枯萎病菌,梨黑星病菌,烟草赤星病菌,番茄灰霉病菌均具有强颉颃作用的芽孢杆菌B-Q2.单孢颉颃性筛选表明B-Q2-7的抑菌活性最强,对稻瘟病菌和番茄灰霉病菌的抑制率分别达到87.4%和90.6%.盆栽实验结果显示B-Q2-7无菌发酵滤液对番茄灰霉病具有良好的预防和治疗作用,10倍液的预防效果和治疗效果分别为89.6%和80.4%.抑菌机理研究表明,B-Q2-7代谢产物对梨黑星和番茄灰霉病菌的孢子萌发具有极强的抑制作用,在稀释10倍时的抑制萌发率分别达到95.9%和100%;显微观察到被B-Q2-7抑制的病原真菌菌丝大部分扭曲、肿大、畸形,出现膨大的泡囊.

Abstract:There were 142 bacillus strains isolated from soil,which collected in the village of Dazhai,Yangling,Shaanxi province.By plate test and cell-free fermentation filtrate screening,B-Q2 was obtained which is a stronger antagonistic bacillus and can inhibit Magnaporthe grisea,Fusarium graminearum,Fusarium oxysporum,Venturia nashicola,Alternaria alternate and Botrytisci nerea.The screening of single-spore inhibiting activity showed that the inhibiting activity of B-Q2-7 was the best,the inhibition rate on Magnaporthe grisea and Botrytisci nerea could reach 87.4% and 91.6% respectively.Results of pot experiment showed that cell-free fermentation filtrate of B-Q2-7 had strong preventing and cure efficacy on Botrytisci nerea,the preventing and cure effect could get to 89.6% and 80.4% respectively when fermentation filtrate was diluted 100 times.Study on inhibiting mechanism of strain B-Q2-7 demonst rated that metabolites produced by antagonistic strain B-Q2-7 could strongly inhibit the sporus germination of Venturia nashicola and Botrytisci nerea.It''s inhibition rate of germination could reach 95.9% and 100% when fermentation filtrate was diluted 100 times.Oberserving by microscope showed that the mycelium inhibited by strainB-Q2-7 mostly twisted,inflated,deformed and formed expanded vacuole.
