高羊茅辐射敏感性和辐照处理对其成熟种子愈伤诱导的影响

摘    要:无菌培养条件下发芽率、成苗率、苗高和根长的测定结果表明 ,4个高羊茅品种的辐射敏感性差异很大 ,美洲虎 3号和可奇思对辐射比较敏感 ,半致死剂量大约在 1 5 0Gy左右 ,凌志和猎狗 5号的辐射敏感性分别中等和较弱。用低剂量γ射线处理可促进高羊茅成熟种子的愈伤形成 ,而且诱导愈伤中胚性愈伤的比例有一定提高。因此 ,成熟种子辐照处理可作为提高高羊茅组培效率的一种辅助手段。

Abstract:Measurement of seed germination, seedling survival and height, and root length under the condition of sterile culture indicated that four tall fescue cultivars varied greatly in radiation sensitivity. Jaguar 3 and Cochise were sensitive to radiation with a median lethal dose (LD 50 ) of about 150Gy, whereas Barlexas and Houndog 5 had moderate and weak radiation sensitivity respectively. Callus induction from mature seeds of tall fescue was stimulated and the ratio of embryogenic calli to induced calli increased to a certain extent by treatment with low dose γ-rays. It was proposed, therefore, that irradiation treatment of mature seeds could be used as a supplementary means of enhancing tissue culture efficiency of tall fescue.
