乌头总碱的纯化工艺及中乌头碱的纯化研究

摘    要:目的]选出乌头总碱的最佳提取工艺,并分离纯化中乌头碱,为新药研制奠定基础。方法]以总生物碱提取率为指标考察提取工艺。通过硅胶柱层析和结晶纯化获得纯度在95%以上的中乌头碱。结果]附子中乌头总碱的最佳提取条件为:以3倍药材量的95%乙醇溶液为溶剂,浸提3次,第1次5 d,第2次3 d,第3次1 d。调提取液pH值为3.0~4.0后静置12 h,回收乙醇并浓缩至一定体积,确定D-101的最佳吸附率、洗脱率分别为87.5%和95.3%。纯化时,D-101大孔吸附树脂的上样浓度为6 g/L,pH值为4.5,室温为18℃,以蒸馏水除杂,流速为1 ml/min的95%乙醇洗脱,浓缩洗脱液,完全蒸干,测得其附子总生物碱的平均含量高于48%。结论]D-101大孔吸附树脂可用于工业化乌头总碱的提取,以及后续各种生物碱的纯化。

Abstract:Objective] The aim was to select out the optimum extraction technology of total aconitines and isolate and purify mesaconitine so as to lay a foundation for preparing new medicines.Method] With the extraction rate of total alkaloid as index,the extraction technology was studied.The mesaconitine with purity higher than 95% was obtained through silica gel column chromatography and crystallization purification.Result] The optimum extraction condition of total aconitines from aconite was as follows: with 3 times amount of 95% ethanol as solvent,extraction for 3 times and for 5 d at the 1st,3 d at the 2nd and 1 d at the 3rd.The extracts was stationary for 12 h since its pH value was adjusted to be 3.0~4.0,the ethanol was recycled and concentrated to a certain volume,the optimum adsorption and elution rates of D-101 were ascertained to be 87.5% and 95.3%.During purification,the sample concn.of D-101 macroporous adsorption resin was 6 g/L,the pH value was 4.5,the room temperature was 18 ℃,the impurities were removed by distilled water,the sample was eluted by 95% ethanol with flow velocity of 1 ml/min,the eluent was concentrated,till dried out completely and the determined average content of total alkaloid from aconite was higher than 48%.Conclusion] D-101 macroporous adsorption resin could be used in the industrial extraction of total aconitines and subsequent purifications of various alkaloids.
