| 摘 要： | 为了明确苏云金芽孢杆菌在河北省不同生态区的分布特点和cry基因的多样性,研究从河北省不同生态地区(阜平天生桥、涞源白石山、安新白洋淀、保定郊区大田和安国中草药田)采集土样806份,采用温度-抗生素法分离获得Bt菌株46株,利用PCR-RFLP技术对46株Bt进行了基因型研究,结果表明从这些菌株中发现7种不同的基因型cry1、cry2、cry3、cry4、cry8、cry30和cry32,11株未鉴定出基因型;通过SDS-PAGE分析发现这些菌株主要表达130-150kDa、70-80kDa、60kDa和30kDa蛋白,实验结果说明了苏云金芽孢杆菌在河北省不同生态区均有分布,并且其cry基因类型是复杂多样的。

| Abstract: | To study distribution of Bacillus thuringiensis and diversity of their cry gene in different ecological area,806 soil samples were collected from Tianshengqiao in Fuping,Baishi Moutaion in Laiyuan,Baiyang Lake in Anxin,crop field in suburb of Baoding,herbs field in Anguo of Hebei.Forty six strains were isolated by temperature-antibiotic method.Seven kinds of cry gene,cry1,cry2,cry3,cry4,cry8,cry30 and cry32,were found by using PCR-RFLP,11 Bt strains harbour unknown cry genes.According to SDS-PAGE analysis,results show that those Bt strains mainly expressed 130-150 kDa,70-80 kDa,60 kDa,30 kDa protein.Theses results demonstrated that Bt widely distributed in different ecological region of Hebei Province and their cry gene type was complex and various. |
