摘    要:为预防菜用大黄玻璃化苗的产生、提高组培苗的培养质量,以菜用大黄丛生芽为外植体,以1/2 MS为基本培养基,研究了蔗糖、琼脂、6-BA对菜用大黄组培苗玻璃化现象的影响,同时测定了玻璃化苗与正常苗的生理指标差异。结果表明:适于菜用大黄组培苗最大增殖及玻璃化率最小的培养基组合是添加30g·L~(-1)蔗糖+6g·L~(-1)琼脂+1.0mg·L~(-1) 6-BA+1.0mg·L~(-1) NAA的1/2 MS培养基;正常苗的可溶性糖和可溶性蛋白含量明显高于玻璃化苗,而玻璃化苗的组织含水量比正常苗的略高。

Abstract:In order to prevent vitrification seedling of Rheum rhaponticum L.,and improve the quality of somaclone,taking Rheum rhaponticum L.buds as explants,with 1/2MS as the basic culture medium,the effects of sucrose,agar and 6-BA on the vitrification of tissue cultured seedlings were studied.Meanwhile,physiological and biochemical indexes between vitrification shoots and normal seedlings were determined and compared.The results showed that the high coefficient of proliferation and the low rate of vitrification was cultured in 1/2 MS medium supplemented with 30 g·L-1 sucrose,6 g·L-1 agar,1.0 mg·L-1 6-BA and 1.0 mg·L-1 NAA.The soluble sugar and soluble protein content of the normal seedlings was significantly higher than that of the vitrification shoots,while the water content of the tissue was slightly higher than that of the normal seedlings.
