棉花亲环素基因(GhCYP1)克隆及在干旱胁迫下的表达分析

摘    要:本研究从已构建的棉花(Gossypium hirsutum)干旱胁迫抑制性差减杂交(suppressive subtractive hybridization,SSH)cDNA文库分离得到一个含有植物亲环素保守结构域的EST片段,测序并结合cDNA末端快速扩增(5'-RACE)技术获得了1个779bp的cDNA序列。序列分析表明,该cDNA5'非翻译区为70bp,3'非翻译区为187bp,并含有一个编码174个氨基酸蛋白的开放阅读框。Blast分析表明,该基因的编码产物为一个亲环素蛋白,将该基因命名为为GhCYP1,序列提交到GenBank,登录号为GQ292530。半定量RT-PCR分析表明,干旱胁迫处理后,该基因在叶片中的表达量迅速提高,并在胁迫2h达到最高,这一研究暗示该基因的表达与棉花抗旱胁迫相关。

Abstract:In this research,we isolated a EST fragment containing the conserved domain of plant cyclophilin from the SSH cDNA library formerly constructed under drought-stress from Gossypium hirsutum L.,then a 779 bp cDNA fragment were obtained by using sequencing and rapid amplification of cDNA ends (5'-RACE).Sequence analysis showed the cDNA contained a 70 bp 5' untranslated region,a 187 bp 3' untranslated region,and an open reading frame encodes 174 amino acids.Blast analysis indicated that its encoded product was a cyclophilin protein,and it named as GhCYP1 with GenBank accession number GQ292530.Semi-quantitative RT-PCR analysis showed that the gene expression levels of GhCYP1 rapidly elevated in the cotton leaves under drought-stress condition,and it reached the highest point after two hours.This study implies that GhCYP1 gene expression related to drought tolerance in cotton.
