摘    要:目的]快速、准确测定米根霉富马酸发酵液中的富马酸含量。方法]在柱温65℃、流动相为5 mmol/L硫酸水溶液、流速为0.8ml/min的色谱条件下,采用离子排斥液相色谱法测定米根霉富马酸发酵液中的富马酸含量。结果]富马酸的保留时间为10.450 min,其他杂质不干扰测定。离子排斥液相色谱法在富马酸质量浓度为0.5~5.0 g/L时具有良好的线性关系,相对标准偏差为0.667%,回收率达98%以上。发酵时间分别为12、487、2 h的米根霉富马酸发酵液中的富马酸的质量浓度分别为5.712、36.4205、6.449 g/L,RSD分别为1.040%0、.667%和0.430%。结论]离子排斥液相色谱法测定米根霉富马酸发酵液中的富马酸含量的速度快,重现性好,准确度高,结果可靠。

Abstract:Objective] The aim was to correctly determine the content of fumaric acid in fermentation broth of Rhizopus oryzae(R.oryzae).Method] Under the chromatogram conditions of column temperature of 65 ℃,mobile phase 5 mmol/L of sulphuric acid solution and velocity of 0.8 ml/min,the ion-exclusion liquid chromatography was used to determine the content of fumaric acid in fermentation broth of R.oryzae.Result] The retention time of fumaric acid was 10.450 min,the other impurities did not interfere determination,the ion-exclusion liquid chromatography had good linear relation when the mass concentration of fumaric acid was 0.5-5.0 g/L,the relative standard deviation was 0.667%,and the recovery was over 98%.When the fermenting time was 12,48 and 72 h resp.,the concentration of fumaric acid in fermentation broth of R.oryzae was 5.712,36.420,56.449 g/L resp.,and RSD was 1.040%,0.667% and 0.430% resp.Conclusion] The ionexclusion liquid chromatography could determine the content of fumaric acid in fermentation broth of R.oryzae rapidly,with good reproducibility,high accuracy and reliable result.
