摘    要:为了解和掌握广西乌榄的资源类型及其遗传变化规律,利用SCoT分子标记技术,对桂西南地区收集到的42份乌榄种质资源进行遗传多样性分析。筛选出14条引物对供试材料进行PCR扩增,共获得124条带(DNA位点),其中具有多态性条带74条,平均多态性比例(PPB)59.68%;经Ntedit软件计算,其遗传相似系数(GS)的范围值在0.726~0.903,平均为0.817,表明SCoT能够揭示乌榄种质间的遗传变异。UPGMA法聚类分析结果显示,在遗传相似系数为0.812时,可将乌榄种质资源分为A、B 2个大组,每大组又各分2个亚组,亚组内种质资源按照采样地聚集,说明乌榄的亲缘关系与地理起源有一定的相关性。总体上,橄榄种质资源遗传多样性水平相对较低。

Abstract:In order to learn and master the resources and genetic change rules of Canarium pimela in Guangxi,genetic diversity of 42 C.pinela germplasm resources was collected in southwest regions of Guangxi and was analyzed by using SCoT markers.The samples were amplified by using 14 selected primers,and 124 bands were obtained,74 of which were polymorphic loci with a polymorphism rate of 59.68%.The similarity coefficient between different germplasms varied from 0.726 to 0.903 with an average of 0.817.The results of cluster analysis by using UPGMA method showed that 42 C.pimela germplasms resources could be clustered into A and B large groups at similarity coefficient of 0.812.Each large group was divided into 2 subgroups,The genetic relationship of germplasm resources in the subgroups had a certain correlation with geographical origin.The genetic diversity of the C.pimela germplasm resources was lower.
