不同饲粮类型对内蒙古绒山羊瘤胃细菌多样性的影响

摘    要:研究不同饲粮类型对内蒙古绒山羊瘤胃液细菌多样性及群落结构的影响。选择24只健康的8月龄、体重约25 kg的内蒙古绒山羊母羊,随机分为4组,每组6只,分别为苜蓿组(G1)、苜蓿补饲精料组(G2)、玉米秸秆组(G3)和玉米秸秆补饲精料组(G4),试验期共25 d,其中预饲期20 d。在正式试验期第5天,经口腔采集瘤胃液,利用高通量测序技术进行瘤胃细菌多样性分析。结果表明:基于相似性>97%的原则,4组样品共产生1 399个操作分类单元(OTU),共享OTU 1 040个,约占总数的74.34%;各组α多样性指数指标无显著性差异(P>0.05)。各组门水平最优势菌为拟杆菌门,科水平最优势菌为普雷沃氏菌科,属水平最优势菌为普雷沃氏菌属,在所有占总细菌比例大于1%的瘤胃细菌中,G2组软壁菌门比例显著高于G3和G4组(P<0.05),G1组毛螺旋菌科比例显著(P<0.05)高于其他3组,其余各组菌群比例均无显著差异(P>0.05)。综合得出,不同饲粮类型条件下,内蒙古绒山羊最优势菌门为拟杆菌门,最优势菌科是普雷沃氏菌科,最优势菌属是普雷沃氏菌属;不同饲粮类型对内蒙古绒山羊瘤胃菌群结构影响不明显。

Effects of different diet types on the rumen bacterial diversity of Inner Mongolia cashmere goats

Abstract:This study investigated the effects of different diets composition on the diversity of bacteria in rumen fluid and their community structure in Inner Mongolia cashmere goats. Twenty-four healthy 8-month-old Inner Mongolia cashmere goats were selected and randomly divided into four groups(n=6), which were alfalfa hay group(G1), alfalfa hay plus concentrate group(G2), cornstalk group(G3)and cornstalk plus concentrate group(G4). The trial period lasted 25 days, of which the pre-test period was 20 days. On the fifth day after the beginning of the experiment, rumen fluid was collected through the mouth and the diversity of rumen bacteria was analyzed using high-throughput sequencing technology. The results showed as follows: At the level of 97% similarity, among the 1 399 operational taxonomic units(OTU)generated in the four groups, 1 040 OTU were shared, accounting for 74. 34% of the total OTU. There was no significant difference in alpha indexes of different diet groups(P>0. 05). The predominant phylum of rumen bacteria in different diet groups was Bacteroidetes, and the predominant bacteria family was Prevotellaceae, and the predominant bacteria genus was Prevotella. Among all rumen bacteria that accounted higher than 1%, The proportion of Tenericutes in group G2 was significantly higher than that in groups G3 and G4(P<0. 05), The proportion of Veillonellaceae in group G1 was significantly higher than that in groups G2, G3 and G4(P<0. 05). Other groups displayed no significant difference in the proportion of bacterial community(P>0. 05). Collectively, the predominant phylum, family, and genus of rumen bacteria were Bacteroidetes, Prevotellaceae, and Prevotella, respectively. No significance was observed among different groups.
