枣幼胚培养中褐化的研究

摘    要:研究了建立枣幼胚离体培养体系时品种、外植体、活性炭、接种方式和基本培养基对褐化的影响,其中,外植体是枣幼胚离体培养时褐化的关键因素。取出早脆王胚珠,剥去珠被接种可以有效防止褐化。不同发育时期的胚珠褐化不同,其结果与品种及接种方法有关。完整接种的早脆王胚珠随其成熟度的增加褐化加重,但切开远胚端接种金丝丰和冬枣的幼嫩胚珠更容易褐化。加倍的MS培养基可以促进早脆王幼胚褐化,附加活性炭可以减轻早脆王完整胚珠褐化。

Abstract:Studies on factors affecting browning of immature embryo culture of Chinese jujube were carried out,including cultivar, explant, carbo activatus, inoculation method and basic medium, of which explant was the key factor. The ovule browning of Zaocuiwang could be efficiently prevented by removing integument during inoculation. The browning of ovule during different development stage was related to cultivars and inoculation method. While the whole ovule browning of Zaocuiwang was aggravated with maturation, the ovules browning of Jinsifeng and Dongzao with the end dissection far away from the embryo were easier. The immature embryo browning of Zaocuiwang was accelerated on double MS medium and the whole ovule browning of Zaocuiwang could be alleviated through addition of carbo activatus.
